金黄色葡萄球菌肠毒素B对体外培养小鼠胚胎发育的影响

摘    要:目的了解金黄色葡萄球菌肠毒素B(SEB)对胚胎发育的影响。方法采用植入后全胚胎培养技术,将8.5d小鼠胚胎与终浓度分别为0.1、1及10μg/ml的SEB共培养48h后,观察SEB对胚胎生长发育和器官形态分化影响。结果SEB可影响体外小鼠胚胎的发育。0.1μg/mlSEB首先影响卵黄囊直径,当SEB为1及10μg/ml时,胚胎生长发育指标和器官形态分化评分均明显低于对照组,并出现小头、脑膨出、前后神经管未闭、体位异常、鳃弓及前肢发育迟缓、大心包及心包积液等畸形。结论SEB能明显影响体外小鼠胚胎发育,可能是金葡菌L型导致体外胚胎发育畸形的主要机制之一。

Abstract:In order to understand the influence of staphylococcal enterotoxin B (SEB) on the embryonic development of mice, a post-implantation technique of the whole embryo culture (WEC) was used in which the embryonic development and morphological differentiation were observed 8.5 days after co-cultivation of mouse embryos with 0.1, 1.0 and 10 g of SEB for 48 hours. It was observed that the embryonic development of mice could be damaged by SEB in vitro. The diameter of yolk sac was firstly affected by 0.1 g of SEB and the indices of embryonic development and the morphological scores of mice co-cultured with 1.0 and 10 g of SEB were significantly lower than that of control with morphological and developmental abnormalities. In conclusion, it is apparent that SEB can interfere with the development of mouse embryos as demonstrated in vitro, and this effect may be one of the mechanism of teratogenieity of the cultured embryos induced by the L form of Staphylococctis aureus.
