脂质体对羟喜树碱内酯环的保护

摘    要: 目的考察脂质体对羟喜树碱的结构中α-羟基内酯环的保护作用。方法采用HPLC测定模拟生理pH下(pH7.4磷酸盐缓冲液,37℃)羟喜树碱脂质体中羟喜树碱以内酯环结构存在的含量。色谱条件为:色谱柱为C18柱(5μm,4.6 mm×250 mm),流动相为甲醇-水(60:40),检测波长为266 nm。结果在模拟生理pH下24 h受脂质体包裹的羟喜树碱的闭环率仍大于90%。结论说明脂质体对羟喜树碱结构具有较好的保护作用,能有效地保护羟喜树碱的α-羟基内酯环。

Abstract:OBJECTIVE To investigate the potential of liposome for stabilizing lactone ring of hydroxycampothecin(HCPT) . METHODS HPLC method was used to determine lactone form of HCPT in liposomes in stimulated physiological environment. HPLC method was performed on a C18 column (5μm,4.6mm×250 mm) with the mobile phase of methanol-water(60:40). HCPT was determined at 266 nm. RESULTS More than 90% HCPT in liposomes remained its lactone form in stimulated physiological environment for 24 h. CONCLUSION Liposomes may be a useful dosage form to stabilize HCPT.
