摘    要:目的探讨一种改良的大鼠海马透射电镜(以下简称电镜)快速取材方法。方法16只SD大鼠被随机分为常规灌注固定取材组(常规组)和改良取材组(改良组)。改良组从动物断头处死、颅骨剥离及海马取出方式等操作方面进行了改进,对2组取材时间和取材效果进行比较。结果改良组和常规组平均取材时间分别为(1.27±0.07)min、(21.67±0.88)min,改良组明显短于常规组(P<0.01)。电镜下观察2种取材方法均保证了大鼠海马超微结构完整无损伤。结论改良法缩短了取材时间,提高了取材效率,尤其适用于大批样本量的取材。

Abstract:Objective To investigate an improved transmission electron microscope (TEM) sampling, method of rat hippocampus. Methods Sixteen SD rats were randomly divided into convention group and improvement group. The experimental procedures were improved in sacrifice of animal, skull gaffing and the way of taking out of hippocampus. The sampling time and effect were compared between two groups. Results The sampling time in improvement group and convention group were( 1.27 ± 0.07 ) min and (21.67 ±0.88 )min respectively, the sampling time in improvement group was shorter than that in convention group( P 〈 0.01 ). The uhramicrostructures of hippocampus were complete and scatheless in two groups. Conclusion The improved method could shorten the sampling time and increase the sampling rate, especially suitable for large sampling.
