摘    要:目的:采用大鼠盲肠结扎穿孔(CLP)复制脓毒症模型造成急性肝损伤,探讨自由基代谢在脓毒症肝损伤发病机制中的作用。方法:将Wistar大鼠随机分为对照组、假手术组和模型组,每组叉分6h、12h、24h和48h四个时间点。动态观察血清丙氨酸转氨酶(ALT)、血清和肝组织中丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH—Px)的变化。结果:从血清含量变化和肝脏细胞形态学检测确认CLP脓毒症模型造成了肝损伤。CLP组血清和肝组织MDA于6h时间点增高,48h达到峰值,血清和肝组织GSH—Px活性亦于6h时间点降低,48h达到最低值。结论:脓毒症所致的肝细胞损伤与自由基代谢紊乱有关。

Abstract:Objectives: To investigate the role of free radical metabolism in septic liver injury on the model of sepsis in rat induced by cecal ligation and puncture.Methods: The male rats were assigned randomly to control group(Control),sham operated group(Sham),cecal ligation and puncture group(CLP).The levels of alanine transaminase(ALT) and serum levels of malondialdyhyde(MDA) and serum and hepatic tissue level of glutathione peroxidase(GSH-Px) were examined consecutively at 6h,12h,24h,and 48h.Results: Septic liver injury was affirmed by the changes of serum ALT levels and hepatic morphocytology.Compared with Sham group,MDA levels both in serum and hepatic tissue in CLP group began to increase at 6 hour and reached their peak at 48 hour.GSH-Px level both in serum and hepatic tissue in CLP group began to decrease at 6 hour and reached their minimum at 48 hour.Conclusions: Disturbance of free radical metabolism may be related to the mechanism of liver injury in sepsis.
