哮喘小鼠Galectin-8和MMP-9的表达及布地奈德的干预作用

摘    要:目的:探讨哮喘小鼠气道和肺组织中半乳糖凝集素8(Galectin-8)及基质金属蛋白酶9(MMP-9)的表达情况及布地奈德对其表达的影响。方法:36只清洁级雌性BALB/c小鼠随机分为对照组、哮喘组、布地奈德干预组,建立卵清蛋白(OVA)致敏、激发的哮喘模型,观察各组小鼠肺组织的病理变化;免疫组化、RT-PCR分别测定各组小鼠气道和肺组织中Galectin-8、MMP-9蛋白和mRNA的表达;结果 :哮喘组小鼠气道和肺组织中Galectin-8、MMP-9蛋白和mRNA的表达显著高于对照组及布地奈德干预组(P<0.05),Galectin-8蛋白和MMP-9蛋白的表达呈正相关(r=0.89,P<0.05)。结论:哮喘小鼠气道和肺组织中Galectin-8、MMP-9蛋白和mRNA的表达明显升高,并且两者蛋白的表达具有相关性;布地奈德干预后Galectin-8、MMP-9蛋白和mRNA表达下调。

Abstract:Objective To study the expression of Galectin-8 and MMP-9 in airways and lungs of mice with asthma and the effect of budesonide on the expression.Methods Thity-six BALB/c SPF female mice were randomized into control group,asthma group and budesonide intervention group.Establish a mouse asthma model by sensitization and challenge with ovalbumin(OVA),observe the pathological changes of the lung tissues in all mice,the expression of Galectin-8 and MMP-9 protein were detected by immunohistochemistry,Galectin-8 and MMP-9 mRNA levels in lung tissues were determined using RT-PCR.Results The protein and mRNA expression of Galectin-8 and MMP-9 in airway and lungs of mice in asthma group significantly high than the mice in control group and budesonide intervention group.(P<0.05).There was positive correlation between the protein expression of Galectin-8 and MMP-9(r=0.89,P<0.05).Conclusion The protein and mRNA expression of Galectin-8 and MMP-9 in airway and lungs of mice with asthma significantly increased,there was positive correlation between the protein expression of Galectin-8 and MMP-9.The protein and mRNA expression of Galectin-8 and MMP-9 in airway and lungs of mice decreased after budesonide treatment.
