Discussion on the mechanism of Fuzhen Kang'ai formula in the treatment of liver cancer based on network pharmacology

Objective To study the effective components,targets and possible mechanisms of Fuzhen Kang'ai formula in the treatment of liver cancer based on the method of network pharmacology.Methods The bioactive components of Fuzheng Kang'ai formula were screened by using the traditional Chinese medicine system pharmacology database and analysis platform(TCMSP),and their targets were predicted.The pharmacodynamic molecule and target network was constructed,the protein interaction was analyzed,and the potential targets were further found.Finally,the possible molecular mechanism was analyzed by KEGG pathway and GO enrichment.Results Two hundred and fourteen active components and 260 molecular target genes were obtained from TCMSP.PPI analysis further obtained 4875 indirect targets,and 111 common targets were obtained by intersection with 534 liver cancer targets.KEGG enrichment analysis showed that the targets of Fuzhen Kang'ai formula were enriched in the pathways related to the occurrence and development of liver cancer,such as cancer pathways,cell cycle proliferation,DNA synthesis and other pathways.GO functional analysis showed that these targets were mainly involved in cell proliferation,DNA synthesis,damage repair and gene expression.At the same time,these targets were mainly distributed in the cytoplasm and membrane,facilitating the binding of protein and ATP,and were good potential drug targets.Conclusion The Fuzhen Kang'ai formula may mainly treat liver cancer by interacting with key target proteins such as PTGS2,NCOA2,ESR1,MAPK1/3,TP53 and TNF,inhibiting inflammatory response,regulating immune escape,inhibiting liver cancer cell proliferation and metastasis,blocking cell proliferation cycle and inducing apoptosis.
