Prediction of related targets/pathways of Ganmai Dazhao decoction in the treatment of postpartum depression

Abstract:Objective To explore the action target of Ganmai Dazhao decoction in the treatment of postpartum depression by network pharmacology method, and to clarify its related mechanism. Methods The chemical components of Ganmai Dazhao decoction were collected by traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP), and the active components and action targets were screened. The targets related to postpartum depression were retrieved through GeneCards database. The common targets of drug and disease were screened and collected by Venny 2.1 software. The common targets of drug and disease were input into the String database to obtain the protein-protein interaction(PPI) network diagram, and then the core targets were analyzed by Cytoscape 3.7.2 software. Finally, GO and KEGG enrichment analysis were performed by DAVID database. Results A total of 119 active components and 254 corresponding targets of Ganmai Dazhao decoction were obtained by screening;315 targets related to postpartum depression with Score value ≥5 were collected;there were 62 potential targets related to postpartum depression in Ganmai Dazao decoction by target mapping, mainly involving SCN5A, PTGS2, HTR3A, NOS2, ESR1,MAPK14, DRD1 and so on. The enrichment of GO entry targets mainly focused on biological processes(BP), involving system function, multicellular biological process, molecular function(MF) and so on;the targets were mainly distributed in extracellular space, extracellular region, membrane raft, cell membrane and so on;the effects of targets included amine binding, amine receptor binding, cytokine activity, drug binding and so on. The results of KEGG enrichment showed that the main targets of Ganmai Dazhao decoction in the treatment of postpartum depression were related to the signaling pathways such as intestinal immune network regulating IgA, neuroactive ligand-receptor, cytokines, cytokine receptors and so on. Conclusion This paper systematically explains the mechanism of Ganmai Dazhao decoction in the treatment of postpartum depression through the analysis of network pharmacology, which provides a theoretical basis and new ideas for the further treatment of postpartum depression.
