Analysis of the Effect Mechanism of Modified Chaihu Longgu Oyster Decoction in the Treatment of Insomnia Based on Network Pharmacology

Abstract:Objective]To study the mechanism of modified Chaihu Longgu Oyster decoction in the treatment of insomnia based on network pharmacology technology.Methods]Through Traditional Chinese Medicine System Pharmacology Analysis Platform(TCMSP) by setting OB≥30%, DL≥0.18 and searching BATMAN database, the active ingredients were found in the modified Chaihu Longgu Oyster decoction, and the potential targets were screened by analysis and prediction through Swiss Target Prediction platform;the insomnia related targets were obtained by searching the databases as OMIM, DiGSeE and GeneCard, and component-target and the other multi-level network association graph were constructed by Cytoscape software. The GO and KEGG enrichment analysis were conducted for the effective targets by DAVID database.Results]A total of 203 active components were screened out by TCMSP and BATMAN, and 161 were obtained after removing the duplicates. According to the OB value and DL value,the top 30 herbal and mineral components were selected for analysis, and a total of 302 potential targets were found. In total of 98 effective targets from the modified Chaihu Longgu Oyster decoction were found after the 2 620 insomnia targets from the disease database intersected with the potential targets in the form of Venny diagram. GO and KEGG enrichment analysis was conducted on the effective targets by DAVID database, and the enrichment results with P<0.05 were selected for analysis according to the p-values from small to large.Conclusion]In this study, it was analyzed for the multi-component, multi-target, multi-pathway of modifed Chaihu Longgu Oyster decoction and its related components-target network effect diagram. It provides the data network support and thus lays a foundation for the later experimental researches in the treatment of insomnia with modifed Chaihu Longgu Oyster decoction.
