细胞间粘附分子-1水平与胃癌浸润和转移相关性的研究

目的研究胃癌患者胃癌组织及外周血中细胞间粘附分子-1(ICAM-1)与胃癌浸润和转移能力的相关性。方法通过采用免疫组织化学染色和流式细胞仪分别检测胃癌患者胃癌组织和外周血中ICAM-1的水平。结果ICAM-1在胃癌组中腺癌和粘液细胞癌均有表达,其中高分化腺癌表达水平高于低分化腺癌,有肝脏转移癌组高于无转移组。胃溃疡组织中未见表达。可溶性细胞间粘附分子-1(sICAM-1)在胃癌患者血清水平明显高于正常对照组,有肝脏转移癌组明显高于无转移癌组。结论ICAM-1在胃癌组织中表达可能有助于肿瘤细胞浸润和转移的发生。而ICAM-1作为免疫抑制因子阻断了ICAM-1/LFA-1系统功能,削弱了机体免疫监视能力和对肿瘤细胞的破坏作用,导致肿瘤细胞的不断生长和发生远处转移。

Study about the correlation of the level of ICAM-1 and the invasiveness and metastasis in gastric carcinoma

Objective To investigate the correlation of the level of ICAM-1 and the invasiveness and metastasis in gastric carcinoma.Methods Immunohistochemical stain and FCMS were used to determine the level of ICAM-1 in gastric cancer tissues and peripheral blood of gastric cancer patients.Results ICAM-1 expressed in both gastric adenocarcinoma and mucus-cell carcinoma.The expression of ICAM-1 in well-differentiation tissues was significantly higher than that in poor-differentiation tissues.ICAM-1 expression in liver metastasis group was higher than that in non-metastasis group. There was no expression of ICAM-1 in gastric ulcer.The level of ICAM-1 was higher in serum of gastric cancer patients than in the normal group,and was higher in liver metastasis group than in non-metastasis group.Conclusion The expression of ICAM-1 correlated with the invasiveness and metastasis in gastric carcinoma.As an immunological supervision inhibitor,ICAM-1 can block the interaction of ICAM-1/LFA-1,decrease the immunological supervision and cytotoxicity against cancer cells,conduce to the invasiveness and metastasis of gastric carcinoma.
