核酸脉冲电泳场技术在霍乱分子流行病学调查中的应用

摘    要:目的:对深圳市某一霍乱疫点进行流行病学调查和传染来源追踪。方法:采用病原学方法和核酸脉冲电泳场技术,分析霍乱阳性菌株的酶切图谱,并比较图谱的差异。结果:同一疫点的5株霍乱阳性菌株均表现为DNA片段大小一致和数量相同的电脉图谱。结论:5株菌株为同一克隆的流行株。核酸脉冲电泳场分析技术适用于分析菌株的同源性和传染来源追踪。

Abstract:Objective:To make an epidemiologic investigation and look for infection sources.Method:Large restriction fragment(LRF) patter analysis of genomic DNA of cholera positive strains was performed using Pulsed Field Gel Electrophoresis and aetiological method,meanwhile,differences of patterns were made.Results:5 positive strains had the same LRF pattern which showed the same quantity and the same molecular weight of DNA fragments.Conclusion:Five strains were the same cloning epidemic strains.The market was the infectious source of the epidemic locus at the molecular level.So Pulsed Field Gel Electrophoresis was suitable for evaluating homology and looking for infectious sources.
