Zileuton对胃癌SGC-7901细胞侵袭转移的影响

摘    要:目的观察5-脂氧合酶(5-1ipoxygenase,5-LOX)抑制剂Zileuton对胃癌SGC-7901细胞异质黏附、侵袭、迁移、诱导新生血管形成能力的影响,并初步探讨其可能机制。方法用Zileuton处理SGC-7901细胞,MTT法检测其黏附能力的变化;Transwell Chamber观察其侵袭及迁移能力的改变;观察SGC-7901细胞条件培养基诱导内皮细胞形成血管管腔能力;RT-PCR和Western blot检测SGC-7901细胞中骨桥蛋白(osteopontin,OPN)和尿激酶纤溶酶原激活物(urokinase-type plasminogen activator,uPA)的表达变化。结果经6.30×10-5mol.L-1的Zileuton处理后,SGC-7901细胞游走与侵袭穿膜细胞数、诱导内皮细胞形成血管管腔数与管腔长度均低于对照组(P<0.01);黏贴率、OPN和uPA表达均较对照组明显降低(P<0.05)。结论 5-LOX抑制剂Zileuton能有效抑制SGC-7901细胞的侵袭转移,其作用机制可能与抑制OPN和uPA的表达有关。

Abstract:Aim To investigate the effects of Zileuton on the invasion and metastasis of human gastric adenocarcinoma cells SGC-7901 in vitro.Methods The adhesion ability was deteced by MTT,the invasion and metastasis in Transwell Chamber membrane invasion culture system were observed,the capability of SGC-7901 cell condition medium in inducing endothelial cell to form blood vessel was detected,and the expression of OPN and uPA was analysed by RT-PCR and Western blot.Results After treating with Zileuton at concentration of 6.30×10~-5 mol·L~-1 for 72 h,the invading and migrating numbers of SGC-7901 cells,the number and length of vessel were all significantly lower in Zileuton treated groups compared with control groups (P<0.01);the adhesion rate,the expression of OPN and uPA in Zileuton treated groups were all obviously lower than those in controls(P<0.05),respectively.Conclusion Zileuton can inhibit the migration and invasion of SGC-7901 cells in vitro,probably via down-regulating OPN and uPA.
