L—NNA对兔大脑中动脉阻断后脑梗塞灶范围的影响

摘    要:电凝阻断兔大脑中动脉,造成兔局灶脑缺血。于缺血前后静脉注射一氧化氮合成酶抑制剂L—NNA,24h后测量梗塞灶范围并与单纯缺血组比较,发现L—NNA扩大兔大脑中动脉阻断后脑梗塞灶范围。另设一氧化氮合成底物L-精氨酸组等进行对照研究。结果证明该作用是由于减少了一氧化氮的生成而产生。

Abstract:We investigated whether N-nitre-L-arginine(L-NNA),acompetitive inhibitor of nitric oxide synthase(NOS), would modify the volume of the focal ischemic infarction produced by middle cerebral artery occlusion(MCAO) in the rabbit,L-NNA(5mg/Kg) or an equivalent volume of saline was infused into New Zealand white rabbits before and after MCAO. After 24 hours ischemia,the infarction volume in L-NN A -treated animals were increased compared with one of controls (19. 28 2. 26mm2VS 15. 62 2. 56mm2,P<0. 05; mean SD). This effect was antagonized by infusion of L-hut not D - argmine. The findings support the view that L-NNA enhances infarction volume by nitric oxide - dependent mechanisms.
