糙叶败酱提取物的体外抗肿瘤活性比较

摘    要:目的:比较糙叶败酱提取物糙叶败酱粗多糖、糙叶败酱总木脂素、糙叶败酱总皂苷的体外抗肿瘤活性,初步筛选糙叶败酱抗癌作用有效成分。方法:采用四甲基偶氮唑盐比色法(monotetrazolium test,MTT)测定不同浓度糙叶败酱粗多糖、糙叶败酱总木脂素、糙叶败酱总皂苷分别对人肺源腺癌细胞SPCA-1、人肝癌细胞系HepG2、和人慢性髓系白血病细胞K562生长的影响。结果:糙叶败酱总木脂素、总皂苷对体外培养的肿瘤细胞SPCA-1、HepG2、K562均有抑制作用,且抑制作用与药物浓度及作用时间有相关性;糙叶败酱总木脂素对K562细胞生长的抑制作用尤为显著,400μg/ml,作用96h,IC50为21.16μg/ml;糙叶败酱粗多糖对SPCA-1细胞的增殖有抑制作用。结论:糙叶败酱不同成分有不同的抗肿瘤作用,糙叶败酱总木脂素对人慢性髓系白血病细胞生长有明显的抑制作用。

Abstract:Objective:Comparing the differences among Patrinia scabra Bunge.extractions on growth-inhibiteng effects of tumor cell in vitro and testing anti-tumor effects of different Patrinia scabtra Bunge.extractions.Methods:The SPCA-1、HepG2、K562 cells growth was measured by MTT-microculture tetrazoliu assay.Results:Patrinia scabra Bunge.lignanoid、saponin inhibited the SPCA-1、HepG2、K562 cell growth in concentration-and time-dependent manners.And the effect of Patrinia scabra Bunge.lignanoid is more significant.IC50 is 21.16μg/ml after intervention in 400μg/ml Patrinia scabra Bunge.llignanoid for 96h,But polysaccharide has different effect on different tumor cell.Conclusions:The anti-tumor effect of different Patrinia scabra Bunge.extractions are distinguished from treating different tumor cell.The effect of Patrinia scabra Bunge liynanoid is more significant.
