咽炎片中丹皮酚的提取方法及其含量测定

目的:考察不同提取方法对丹皮酚提取率的影响,建立咽炎片中丹皮酚含量测定的方法.方法:对比回流法和超声法进行前处理,重点考察溶剂用量、超声时间、水浴温度等因素对超声法提取率的影响.含量测定采用反相HPLC法,以C18柱分离,甲醇-水(45:55)为流动相,检测波长274 nm.结果:温水浴超声法为咽炎片中丹皮酚含量测定的最佳提取方法,加样平均回收率为99.47%,RSD=0.81%(n=6),含量数据重复性好,RSD=1.04%(n=6).结论:建立咽炎片中丹皮酚的含量测定方法:温水浴超声法是准确性、稳定性、可行性俱佳的方法.

Extraction and Content Determination of Paeonol in Pharyngitis Tablets

Objective: To investigate the effects on the extraction rate of paeonol by different extraction methods and to establish a method for the determination of paeonol in Pharyngitis Tablets.Methods: The extracion rate was compared between the reflux and ultrasonic methods.Among the factors that affect the extraction rates of paeonol,the amount of solvent added,the ultrasonic time,and the bath temperatures were primarily researched.Paeonol was determined by RP-HPLC method,using the column of C₁₈,the mobile phase of methanol-water(45:55),and the detection wavelength of 274 nm.Results: The ultrasonic method in lukewarm water bath is a better pretreatment method,the average recovery was 99.47% with RSD 0.81%(n=6).The results showed a good repeatability with RSD 1.04%(n=6).Conclusion: The ultrasonic procession in lukewarm water bath is a good method for the determination of paeonol in Pharyngitis Tablets due to the high accuracy,stability and practicability.
