高同型半胱氨酸血症对大鼠脑内NMDA受体亚单位蛋白表达的影响

摘    要:目的:分析高同型半胱氨酸血症对大鼠脑内NMDA受体亚单位蛋白表达的影响。方法研究时间2015年7月—2016年4月,将36只SD大鼠随机分为模型组(皮下注射用生理盐水配成的蛋氨酸溶液)和对照组(相应体积的生理盐水),检测不同时间点大鼠血浆同型半胱氨酸含量及脑内NMDA受体亚单位(NR1、NR2A、NR2B)的蛋白含量。结果模型组中第3小组大鼠的Hcy、脑内NMDA受体NR1、NR2A、NR2B分别为(21.46±1.37)、(0.99±0.13)、(0.95±0.18)、(1.47±0.25),均高于第2小组和第1小组(P<0.05),第1小组大鼠上述指标含量均为最低,分别为(10.14±1.13)、(0.74±0.11)、(0.70±0.23)、(0.68±0.13)。模型组3个小组大鼠的脑内NMDA受体NR1、NR2A、NR2B含量均高于对照组中对应小组大鼠,差异有统计学意义(P<0.05)。结论高同型半胱氨酸血症会导致大鼠脑内NMDA受体亚单位的蛋白表达上升,加重大鼠脑神经损伤。

Effects of Hyperhomocysteinemia on NMDA Receptor Subunit Protein Ex-pression in Rat Brain
Abstract:Objective To investigate the effect of hyperhomocysteinemia on the expression of NMDA receptor subunit in rat brain. Methods Study time From July 2015 to April 2016,36 SD rats were randomly divided into a model group (the normal saline solution for subcutaneous injection)and a control group (the corresponding volume of saline),and the rats were sacri-ficed at different time points The plasma levels of homocysteine and the protein content of NMDA receptor subunits (NR1, NR2A, NR2B)in rats were determined. Results The levels of NMDA receptors NR1,NR2A and NR2B in the model group were (21.46±1.37),(0.99±0.13),(0.95±0.18),(1.47±0.25),(10.14±1.13),(0.74±0.11),(0.70±0.23),(0.68±0.13),respectively (P<0.05), and there was no significant difference between the group 1 and group 1.The contents of NMDA receptor NR1,NR2A, NR2B in three groups of rats in the model group were higher than those in the control group(P<0.05) the difference was sca-tistically significant. Conclusion Hyperhomocysteinemia can increase the protein expression of NMDA receptor subunits in the brain of rats and increase the damage of the cranial nerves.
