胶质瘤源性exosome的提取及形态学观察

摘    要:目的证实体内和体外培养的胶质瘤细胞是否均可分泌exosome,为测定其蛋白质组成提供实验基础,为进一步利用exosome对胶质瘤进行免疫治疗提供理论依据。方法收集U251胶质瘤培养上清液和Ⅲ级星形胶质瘤囊液,利用差速离心法提取exosome,并用液相载网法在电镜下观察胶质瘤源性exosome的形态。结果 U251胶质瘤和Ⅲ级星形胶质瘤细胞均可以产生exosome,其平均直径约100 nm。结论证实了胶质瘤细胞像其他多数细胞一样,体内和体外培养均可分泌exosome。

Extraction and morphology observation of exosome from glioma cell

Abstract:Objective To demonstrate whether the glioma cell can secrete exosome both in vivo and in vitro,to provide an experimental basis for the determination of the protein composition,to provide a theoretical basis for immunotherapy for the further use of exosome for glioma.Methods Extracting exosome from hydatid fluid of grade Ⅲ astroglioma cell and culture fluid of U251 glioma cell by differential centrifugation,observing the morphology of glioma cell derived exosome by electron microscope.Results All the U251 glioma cell and grade Ⅲ astroglioma cell can secrete exosome,the average diameter of the exosome is about 100 nm.Conclusion Confirmed that glioma cell,like most other cells,can secrete exosome both in vivo and in vitro.
