核磁共振技术快速检测单纯疱疹病毒Ⅱ型感染细胞的实验研究

摘    要:选用核磁共振技术进行病毒学诊断实验研究结果表明:单纯疱疹病毒Ⅱ型感染细胞后1 h,随着病毒悬液稀释度的不同(10~9~10~(-3)),细胞悬液的自旋晶-格驰豫时间 T_、从11.847 s 相应地缩短至7.807 s,当病毒悬液稀释度为10~(-4)时,细胞悬液的 T_1为12.388 s,恢复到正常对照细胞悬液的 T_1水平(12.175 s)。用不同滴度的抗 HSV-Ⅱ血清中和 HSV-Ⅱ,细胞悬液 T_1相应地逐步恢复至正常。实验研究证实了病毒感染细胞后1 h 就能引起细胞溶液自旋-晶格驰豫时间的特征性改变.运用核磁共振技术可望为临床快速诊断病毒感染提供一条新型、可靠的途径。

Abstract:We make the study on virelogical diagnosis with nuclear magnetic resonance(NMR)technique. The results obtained showed that the Hep-2 cell water proton spinlattice relaxation time(T_1)changed from 7+807 s to 11.847 s in cell cultures with variance dilution of infection(t0~(-3)~10~0)after 1 h adsorption with HSV-Ⅱ preparations at 37℃,the cell control water T_1 was 12.175s,After 1 h ad- sorption with 10~(-4)HSV-Ⅱ preparation at 37℃,the cell Water T_1 was 12.388 s and returned to the level of the cell control water T_1.When HSV-Ⅱ was neutralized by specific anti-HSV-Ⅱ antisorum, it could prevent the change in cell water T_1.The result was similar to the cytopathic effect(CPE).It is confirmed that occured a characteristic change of the cell water T_1 in cell culture after 1 h virus in- fection.These experiments demonstrate that NMR technique is a reproducible,specific,rapidly and easlly method for detection of virusinfceted cell.
