星形胶质细胞条件培养液对机械损伤神经元的保护

摘    要:目的研究体外培养的小鼠星形胶质细胞(Ast)条件培养液(AcM)对机械损伤神经元的保护作用。方法用微量移液器塑料滴头机械性划割培养之神经细胞建立神经元机械损伤模型,神经元培养液加入ACM。用台盼蓝染色计数法测定存活细胞数,测定损伤神经元释放一氧化氮(NO)、乳酸脱氢酶(LDH)的变化,并与常规培养基组进行对比。结果20%ACM可对机械损伤神经元存活增殖起明显促进作用,并减少机械损伤神经元NO、LDH的生成和分泌。结论ACM对机械损伤神经元的存活有促进作用,可减少其NO、LDH的生成和释放,保护机械损伤神经元。

Abstract:Objective To study the effect of Astrocyte conditioned medium(ACM) on mechanical injuried neurons.Methods The neurons mechanical injury model was caused by micropipette tip.The 20% ACM of 5th day of 3rd era were added to the medium of mechanical injuried neurons,then,the surviving cell number was counted by typan blue dyeing;and the concentration of nitric oxide(NO),lactate dehydrogenase(LDH)of the neurons were detected.The data were compared with that of neurons cultured in regular medium.Results Vs control group,20% ACM in medium could obviously promote the survival and proliferation of mechanical injuried neurons,and reduced the production and secretion of NO,LDH.Conclusion ACM can promote mechanical injuried neurons' survival and proliferation,can protect mechanical injuried neurons by reducing the production of NO and LDH.
