MiR-22经上皮-间质转化调控头颈鳞癌的侵袭转移

摘    要:目的探讨miR 22对头颈部鳞状细胞癌(头颈鳞癌)上皮-间质转化(epithelial mesenchymal transition, EMT)和侵袭转移的影响。方法利用脂质体介导的miR 22 mimic及inhibitor(含阴性对照)分别转染头颈鳞癌Tu686细胞,划痕实验及Transwell侵袭实验检测其体外迁移及侵袭能力的改变,Western blotting检测EMT分子标志物的改变。结果hsa miR 22 mimic能抑制Tu686细胞的体外迁移及侵袭能力,相反hsa miR 22 inhibitor能促进Tu686细胞的体外迁移及侵袭能力。且miR 22过表达后能在Tu686细胞中抑制EMT的分子标志物改变,即E cadherin的表达上调和Vimentin的表达下调。结论本研究表明miR 22对头颈鳞癌细胞的体外迁移侵袭能力具有调控作用,该调控作用的发生与miR 22对EMT的抑制作用密切相关。

MiR 22 regulates the invasion and metastasis via remodeling epithelial mesenchymal transition in head and neck squamous cell carcinoma
Abstract:ObjectiveTo investigate the effect of miR 22 on the epithelial mesenchymal transition (EMT), invasion and metastasis in head and neck squamous cell carcinoma (HNSCC) in vitro.MethodsMiR 22 mimic or inhibitor was transfected into cells of head and neck squamous cell carcinoma (Tu686) by liposome transfection technique. Alterations of migration and invasion were assayed by scratches and transwell experiments. Western blotting was used to detect the changes of EMT associated markers.ResultsMiR 22 mimic successfully inhibited migration and invasion of HNSCC Tu686 cell line in vitro. On the contrary, miR 22 inhibitor enhanced the migration and invasion. Meanwhile, miR 22 mimic caused changes of EMT molecular markers including up regulated expression of E cadherin and down regulated expression of Vimentin.ConclusionmiR 22 has a regulatory effect on the migration and invasion of HNSCC in vitro, which is closely related to the inhibitory effect of miR 22 on EMT.
