Abstract:Objective To investigate the effect of ropivacaine on the proliferation,migration,invasion and apoptosis of lung cancer cell A549 through the PI3 K/Akt signaling pathway.Methods The CCK-8 method was performed to determine the proliferation of lung cancer cells A549 in the ropivacaine groups at the concentration of 0,100,200 and 400μg/ml.A549 cells were divided into control group(unadministered)and ropivacaine group(400μg/ml).Transwell cell method was used to analyzed the cell migration and invasion.Cell apoptosis was evaluated by flow cytometry.The protein expression levels of CDK4,MMP-2,Bax,Bcl-2,PI3 K,p-PI3 K,Akt and p-Akt were assessed by Western blot.Results The result of CCK-8 showed that compared with the 0μg/ml ropivacaine group,100,200 and 400μg/ml ropivacaine significantly reduce the cell survival rate of lung cancer cell A549(P<0.05).Transwell chamber method showed that the number of cell migration and cell invasion of A549 cells in the ropivacaine group was significantly lower than that of the control group(P<0.05).The result of flow cytometry showed that the apoptosis rate of A549 cells in the ropivacaine group was dramatically higher than that in the control group(P<0.05).Western blot result showed that,compared with the control group,the protein expression levels of CDK4,MMP-2,Bcl-2,p-PI3 K and p-Akt in A549 cells in the ropivacaine group were significantly reduced,and the Bax protein expression level was significantly increased(P<0.05),While the expression levels of PI3 K and Akt protein did not change significantly.Western blot result showed that,compared with the control group,the protein expression levels of CDK4,MMP-2,Bcl-2,p-PI3 K and p-Akt in A549 cells of the ropivacaine group were greatly reduced,while the protein expression level of Bax was markedly increased(P<0.05).However,there was no significant change in the protein expression levels of PI3 K and Akt.Conclusion Ropivacaine may inhibit the proliferation,migration and invasion of lung cancer cell A549 and promote its apoptosis by inhibiting PI3 K/Akt signaling pathway.
