锂-匹罗卡品癫痫模型中大鼠苔藓纤维发芽机制研究

摘    要:目的:通过观察锂-匹罗卡品癫痫模型中大鼠实验性癫痫发作时的行为、脑电图及病理生理变化,探讨苔藓纤维发芽(MFS)在癫痫中的发病机制。方法:利用锂-匹罗卡品(Li—PC)制作大鼠急性癫痫持续状态(SE)的癫痫模型,造模后7d、14d、28d,进行苔藓纤维发芽评分。结果:模型组大鼠2周时可见MFS穿过齿状回内分子层颗粒细胞层,进入内分子层及CA3区锥体细胞始层黑色颗粒形成,4周时形成连续密集颗粒带,MFS评分显著高于生理盐水对照组(P〈0.05)。结论:急性期SE神经元损伤致苔藓纤维发芽,可能是慢性颞叶癫痫的病理基础。

Abstract:Objective: To explore the pathogenic mechanisms of mossy fiber sprouting in epilepsy by observing the behaviour of experimental epileptic seizure, electroencephalogram (EEG) and the pathophysiological changes of rats in lithium-pilocarpine epilepsy model. Methods: The epilepsy model of the rat acute status epilepticus was made by lithiumpilocarpine (Li-PC) to carry out the scores of mossy fiber sprouting on the 7th day, 14th day and 28th day after the model made successfully. Results:The rats in the model group were showed that mossy fiber sprouting passed through granular cells in inner molecular layer of the dentate gyrus and the black granules were formed in pyramidal cell layer in CA3 area at the 2nd week. Continuous dense granules were formed at the 4th week. The scores of MFS were significantly higher than those in the saline control group ( P 〈 0.05 ). Conclusion: Mossy fiber sprouting induced by the neuronal injury in acute stage may be the pathological base of chronic temporal lobe epilepsy.
