地塞米松对哮喘小鼠支气管RANTES蛋白和mRNA表达的影响

摘    要:目的:探讨地塞米松对哮喘小鼠调节激活正常T细胞表达和分泌细胞因子(RANTES)蛋白和m-RNA表达的影响.方法:将30只二级雄性BALB/C小鼠分为正常小鼠组(A组)、哮喘小鼠组(B组)和地塞米松处理组(C组).以卵清白蛋白(OVA)致敏激发法制备小鼠哮喘模型.采用免疫组化法和原位杂交法测定RANTES蛋白和mRNA在支气管的表达.结果:免疫组化和原位杂交均显示B组支气管RANIES蛋白和mR-NA的表达均显著高于A组(P<0.01),主要表达细胞是上皮细胞,C组支气管RANIES蛋白和mRNA的表达显著低于B组(P<0.01).结论:RANIES参与哮喘发病机制,上皮细胞是主要的表达细胞;地塞米松对哮喘的拮抗治疗作用部分通过抑制RANTES等趋化因子起作用.

Abstract:Objective: To study the effect of dexamethasone on the protein and mRNA expression of RANTES in murine asthma.Methods:30 male BALB/C mice were randomly divided into three groups : the control group (A group) ,asthma group (B group) and dexamethasone treated group ( C group). In the experiment, the mice model of asthma was established with the ovalbumin (OVA) challenge methods. The protein and mRNA expressions of RANTES were detected with im-munohistochemistry methods and in situ hybridization methods. Results: Immunohistochemistry and in situ hybridization showed that the expressions of RANTES protein and RANTES mRNA around the bronchus of group B were significantly elevated than those of group A ( P < 0.01), the epithelial cell was the chief expression cell, the expressions of RANTES protein and RANTES mRNA around the bronchus of group C were significantly lower than those of group B ( P < 0.01) . Conclusion : RANTES is involved in the pathogenesis of murine asthma, the bronchial epithelial cell is the chief expression cell; Glucocorticoid ( dexamethasone) demonstrates anti-inflammatory effects partly by downregulating the expressions of RANTES protein and RANTES mRNA.
