生长抑素类似物诱导人胆管癌细胞株凋亡及对胞内游离钙的影响

目的 探讨生长抑素类似物SMS201－995（SMS）是否通过诱导人胆管癌细胞凋亡而抑制肿瘤生长，以及与细胞内游离钙的关系。方法 采用人胆管癌细胞系SK－ChA-1，应用MMT比色法，Annexin V-FITC技术及激光共聚焦显微镜技术等进行检测和观察。结果 在SMS作用下CK－ChA-1细胞生长受抑制。SMS处理SK－ChA-1细胞后，Annexin V标记的凋亡细胞增多。细胞凋亡的同时细胞内游离钙浓度升高。结论 SMS可能通过诱导SK－ChA-1细胞[Ca^2 ]i依赖性细胞凋亡，抑制胆管癌生长。

Apoptosis of human cholangiocarcinoma cell line induced by somatostatin analogue and its effect on intracellular free calcium in vitro

Objective To investigate whether apoptosis induced by somatostatin analogue (SMS201-995, SMS) will inhibit the growth of human cholangiocarcinoma and the relationship between apoptosis and intracellular free calcium in vitro . Methods MMT assay, Annexin V-FITC and laser scanning confocal microscopy were performed to identify the inhibitory effect of SMS on cell proliferation and the induction of apoptosis in human cholangiocarcinoma SK-ChA-1 cells. Results SMS significantly reduced the growth of SK-ChA-1 cells and increased the number of cells with annexin V-labeling. Meanwhile the intracellular free calcium concentration was increased. Conclusion SMS can inhibit the proliferation activity by inducing the apoptosis mediated by increase of intracellular free calcium in SK-ChA-1 cell line.
