摘    要:对来源于正常小鼠的骨髓内皮细胞经连续传代培养18个月以上。这一培养是在加入经短期培养而获得的内皮细胞条件培养液及GM-CSF,在纤连蛋白覆盖的培养皿上种入低浓度的骨髓基质细胞完成的。在内皮细胞条件培养液和GM-CSF存在的条件下,细胞平均倍增时间为49小时。快速增殖时,细胞先为圆形,以后可变为立方形或梭形。所有培养的细胞与vWF,VCAM-1抗体染色阳性,UEA-1反应阳性,并产生细胞因子。这一骨髓内皮细胞系的建立可用于研究骨髓内皮细胞在造血过程中的作用。

Abstract:Normal murine bone marrow endothelial cells have undergone in long-term culture of serial passages for more than 18 months. This long-term culture was accomplished by using short-term cultured endothelial cell conditioned medium, GM-CSF, a low plating concentration of bone marrow stromal cells and fibronectin-coated dishes. In endothelial cell cpn-ditioned medium and GM-CSF, cells grew with a mean doubling time of 49 hours. When the cells were in a rapid proliferation, they first rounded up and then became cuboidal or spindle shape. All the cultured cells provided strongly positive staining with yon Willebrand factor (vWF), vascular cell adhesion molecule 1 (VCAM-1) antibodies, UEA-1 and elaborated cy-tokines. The establishment of murine bone marrow endothelial ceil line could be useful for studying the role of bone marrow endothelial cells in hematopoiesis.
