Determination of polydatin and resveratrol in Reyanqing Granules by HPLC

Objective To establish a method for the determination of polydatin and resveratrol in Reyanqing Granules.Method The HPLC system consisting the Luna C18(4.6 mm×250 mm,5 μm)column,the mobile phase consisted of Acetonitrile(A)-0.4%(B,V/V)HAc with the gradient elution(0-40min,A18%-57%,B82%-43%),the flow rate was 1.0 mL·min-1,the column temperature was 25 ℃,The UV detector was set at 284 nm.Results The linear response range was 0.031-0.62 mg·mL-1(r=0.9998) and 0.007-0.130 mg·mL-1(r=0.9998),respectively.The average recovery of protocatechuic acid and protocatechualdehyde were 98.8% and 98.3%,respectively,RSD<2.0%.Conclusions The assay demonstrated that the method had adequate and selectivily to measure the concentrations of accurate,repeatable,it can be applied in quantitative determination of polydatin and resveratrol in Reyanqing Granules.
