帕瑞昔布在大鼠局灶性脑缺血再灌注损伤中的神经保护作用

摘    要:目的]探讨帕瑞昔布在大鼠局灶性脑缺血再灌注损伤中的神经保护作用.方法] Sprague-Dawley雄性大鼠36只,随机分为3组:假手术组(SH组)、缺血再灌注组(I/R组)、帕瑞昔布组(PA组).电凝左侧大脑中动脉、夹闭双侧颈总动脉60min以制备脑缺血再灌注模型,再灌注72h时行神经功能缺失评分、2,3,5-氯化三苯四唑(TTC)染色并计算脑梗死容积百分比、Western blot检测缺血侧半影区高迁移率族蛋白1(HMGB1)表达.结果] I/R组神经功能缺失评分、梗死容积百分比明显高于PA组(P〈0.05);与SH组相比,I/R组缺血侧半影区HMGB1表达减少,而PA组较I/R组HMGB1表达减少(P〈0.05).结论] 帕瑞昔布可减轻大鼠局灶性脑缺血再灌注损伤,可能是通过抑制炎症介质表达而发挥神经保护作用.

Neuroprotective Effect of Parecoxib on Focal Cerebral Ischemia/reperfusion Injury in Rats

Abstract:Objective]To explore the neuroprotective effect of parecoxib on focal cerebral ischemia/reper fusion injury in rats. Methods]Thirty-six male Sprague Dawley rats were divided randomly into the sham group(SH group), the ischemia/reperfusion group(I/R group) and the parecoxib group(PA group). The left middle cerebral artery was permanently occluded by bipolar electrical coagulation and both common carotid ar teries were occluded with miniature clips for 60min to establish the cerebral ischemia/reperfusion model. At 72h after reperfusion, the netirological function deficit was scored and 2,3,5-triphenyl tetrazo/ium chloride (TTC) staining was performed. And Western blot was used to detect the expression of high mobility group protein 1 (HMGB1) in ischemic lateral penumbra field. Results]The neurological function deficit score and the percentage of infarction volume in I/R group were significantly higher than those in PA group( P 〈0.05). Compared with SH group, the expression of HMGB1 in ischemic lateral penumbra field of I/R group decreased, and the expression of HMGB1 in PA group was lower than that in I/R group( P 〈70.05). Conclusion] Parecoxib can alleviate local cerebral ischemia/reperfusion injury in rats. It may play a role in the neuro- protective effect through inhibiting the expression of inflammatory mediators.
