Rab22a在乳腺癌中的表达及其对Her-2的影响

目的]检测Rab22a在乳房纤维瘤与乳腺癌中的表达情况,分析它在表皮生长因子受体2（Her-2）、上皮钙粘附素（E-cadherin）、雌激素受体（ER）、孕激素受体（PR）转运与代谢中的作用,以探讨其与乳腺癌分化或转移的关系.方法]取乳房纤维瘤标本40例,乳腺癌标本74例,用免疫组织化学方法检测Rab22a,分析其与乳腺癌转移、病理分级的关系;同时检测Her-2、E-cadherin、ER和PR的表达,分析其与Rab22a的相关性.结果]Rab22a在乳房纤维瘤组织不表达或低表达,在乳腺癌组织中高表达,且两组之间相比具有显著性差异（P〈0.01）;Rab22a在乳腺癌转移组与非转移组中的表达无显著差异（P〉0.05）,在病理学Ⅰ/Ⅱ级组与Ⅲ级组中的表达无显著差异（P〉0.05）;Rab22a在E-cadherin、ER、PR低表达组与高表达组之间无显著统计学差异,但是Rab22a在Her-2低表达组显著高于Her-2高表达组.结论]Rab22a在乳腺癌中高表达,尽管它与肿瘤的转移和分化程度无关,但它可能通过调节Her-2的运输而影响肿瘤细胞的生物活性.

Effect of the Expression of Rab22a on Her-2 in Breast Cancer

Objective]To detect the expression of Rab22a in breast cancer and fibroma and analyze its role in the transport and metabolism of Her-2, E-cadherin, ER, and PR in order to discuss its relationship with the differentiation and metastasis of breast cancer. Methods]The expression of Rab22a in paraffin embedded tis sues of 74 breast cancer and 40 breast fibromas were detected by immunohistochemical method. Their relationship with metastasis and pathological grade of breast cancer was analyzed. Her-2, E-cadherin, ER, and PR in breast cancer were also measured at the same time, and their relationship with Rab22a was analyzed. Results]There was no expression or low expression of Rab22a in breast fibroma, but there was high expression of Rab22a in breast cancer, and there was significant difference between two groups（ P〈0.01）. No significant difference in Rab22a expression was found between metastatic and non-metastatic groups（ P 〉0.05）, pathological grade Ⅰ / Ⅱ group and grade Ⅲ groups（ P 〈0.05）, low expression group of E-cadherin, ER and PR and high expression group of them. The expression of Rab22a in low expression group of Her-2 was higher than that in high expression group of Her-2. Conclusion] Rab22a is over-expressed in breast cancer. It has no relation with the metastasis and differentiation of the tumor, but it may influence the bioactivity of tumor cells by regulating the transport of Her-2.
