阿托伐他汀对实验性变态反应性脑脊髓炎豚鼠脑组织IL-2、IL-6影响的研究

摘    要:目的探讨实验性变态反应性脑脊髓炎(EAE)豚鼠脑组织细胞因子的变化,探讨阿托伐他汀对EAE发病保护作用的免疫调节机制。方法皮下注射粗制碱性髓鞘蛋白(MBP)建立EAE模型。40只豚鼠分成4组:正常对照组、EAE对照组、EAE小剂量组和EAE大剂量组,每组10只,雌雄各半,放射免疫法测定正常对照组、EAE各组脑组织白细胞介素2(IL-2)、白细胞介素6(IL-6)含量。结果EAE对照组、EAE小剂量组和EAE大剂量组脑组织细胞因子IL-2、IL-6水平显著高于正常对照组(P<0.05),EAE高、小剂量组脑组织IL-2、IL-6水平比EAE对照组低(P<0.05),EAE大剂量组IL-2、IL-6水平比EAE小剂量组低(P<0.05)。结论EAE豚鼠存在明显免疫功能紊乱,阿托伐他汀有降低EAE豚鼠脑组织IL-2、IL-6水平作用,其对EAE发病的免疫保护机制可能是通过抑制IL-2、IL-6产生而发挥。

Abstract:Objective To investigate the changes in cytokines of brain tissue in Guinea pig with experimental allergic encephalomyelitis(EAE) and the immunological regulatory mechanisms of atorvastatin.Methods Experimental allergic encephalomyelitis was induced by subcutaneous administration of coarse basic myelin protein in Guinea pig.40 Guinea pigs were assigned to four groups: the normal control group,the EAE control group,low dose atorvastatin EAE group and high dose atorvastatin EAE group.Cytokines(IL-2,IL-6) levels in brain tissue were measured by radioimmunoassay.Results IL-2 and IL-6 levels in the brain tissue of EAE groups were significantly higher than those of normal control group(P<0,05).IL-2 and IL-6 levels in the brain tissue of low and high dose atorvastatin EAE groups were significantly lower than those of EAE control group(P<0.05).IL-2 and IL-6 levels in the brain tissue of high dose atorvastatin EAE groups were significantly lower those of low dose atorvastatin EAE group(P<0.05).Conclusion Inhibition of the production of IL-2 and IL-6 may be the mechanism,by which atorvastatin exerts its immunological treatment role in controlling EAE.
