紫外线对大鼠晶状体泛素mRNA表达水平的影响

摘    要:目的探讨紫外线对体外培养的大鼠晶状体泛素mRNA表达的影响。方法采用实时荧光定量逆转录聚合酶链反应方法(RT-PCR)检测紫外线照射后培养1h,6h、12h、24h和48h的晶状体泛素mRNA的表达水平。结果紫外线照射后泛素mRNA表达水平下降,照射后1h泛素mRNA表达水平最低,之后有所恢复,照射后48h,其mRNA表达水平基本恢复,与对照组相比没有显著性差异。结论紫外线照射可能通过降低大鼠晶状体泛素基因表达而抑制泛素-蛋白酶体途径(UPP)活性,影响晶状体蛋白降解。

Abstract:Objective To study the effect of ultraviolet( UV)- irradiation on the mRNA expressions of ubiquitin in rat lens in vitro. Methods The mRNA expression levels of ubiquitin in rat lens after UV-irradiation were determined by the real-time quantitative RT-PCR ( SYBR Green ) method. Results The results showed that after rat lens were exposed to UV-irradiation, the mRNA expression levels of ubiquitin began to decrease and reached the lowest levels after lhour. The recovery process showed after 48 hours of recover)., the mRNA expression levels reached nearly the normal level, compared with the control group there has been no noticeable difference. However, the mRNA expression level of GAPDH kept unchanged. Conclusion These results suggested that UV-irradiation might inhibit the activity of ubiquitin-proteasome pathway by reducing the gene expression levels of ubiquitin. It might affect the degradation of lens protein.
