链脲佐菌素诱导大鼠糖尿病肾病模型的建立及肾脏病理结构改变

目的:探讨糖尿病大鼠肾脏结构的变化,建立糖尿病肾病大鼠模型。方法:雄性清洁SD大鼠48只,随机分为实验组(DM组,n=24)和对照组(NC组,n=24)。DM组大鼠一次足量腹腔注射链脲佐菌素(STZ)制备糖尿病动物模型,NC组给予等量的枸橼酸-枸橼酸钠缓冲液。分别于注射后1、4、8和12周,检测体重、血糖、24 h尿量、24 h尿蛋白、肾重指数(肾脏重量/体重)和肾脏病理结构的变化。结果:与NC组相比,DM组大鼠血糖明显升高,肾重指数增加(P<0.01);随着病程的延长,24 h尿量、24 h尿蛋白量均持续升高(P<0.01)。病理切片显示,DM组大鼠肾脏重量增加、肾小体肿胀、肾小囊腔缩小、血管球内皮细胞排列紊乱和肾小体基膜增厚等。结论:STZ可诱导糖尿病大鼠肾脏结构和功能受损,成功制备糖尿病肾病大鼠模型。

Pathological structural changes of the kidney in streptozotocin-induced rat model of diabetic nephropathy
