波形蛋白在大鼠肾间质纤维化中的表达及意义

摘    要:目的:探讨波形蛋白(VIM)在肾间质纤维化(RIF)大鼠模型中的表达及意义。方法:将27只SD大鼠随机分为假手术组(S组)、模型组(U组)及丹参酮IIA磺酸钠治疗组(T组),U组和T组采用单侧输尿管结扎术制作肾小管间质纤维化的动物模型,术后第11天处死大鼠,肾组织HE染色及Masson染色后进行图像分析,评定各组肾小管间质损害、肾间质炎性细胞浸润及肾间质纤维化状况;用免疫组织化学方法检测各组大鼠肾间质的VIM的表达。结果:U组肾小管间质损伤指数(TDI)、肾间质炎性细胞浸润指数(TICI)及肾间质纤维化指数(TFI)均明显高于S组(P〈0.01),U组VIM的表达明显高于S组(P〈0.01),T组的上述各指标分别较U组降低(P〈0.05);TDI、TICI、TFI分别与VIM的表达均呈正相关。结论:VIM可能在促炎症反应和促RIF过程中具有一定作用,丹参酮IIA磺酸钠可通过降低VIM在肾小管间质的表达减轻了RIF。

An Experimental Study on Expression of Vimentin in Rat Renal Tubulointerstitial Fibrosis and Its Significance

Abstract:Objective:To study the expression of vimentin(VIM) in renal tubulointerstitial fibrosis(RIF) of rat models and its significance.Methods: RIF models were established by unilateral ureteral obstruction(UUO) method.Twenty-seven normal male Sprague-Dawley rats were randomly divided into 3 groups: sham-operated group(group S),UUO group(group U) and tanshinone(20mg/kg.d) treated group(group T).Rats in groups S and U were given same amount of water instead of drug treatment.All of the rats were killed on the 11st day of the experiment.Sections of renal tissue were stained with HE or Masson methods,and image analysis was carried out to evaluate the damage in renal tubulointerstitium,inflammatory cell infiltrate in renal interstitium,and the fibrosis of renal interstitium.The expression of VIM was determined with immunohistochemistry method.Results: Tubulointerstitial damage index(TDI),tubulointerstitial fibrosis index(TFI) and tubulointerstitial inflammatory cell infiltration index(TICI) in group U were significantly higher than those in group S(P0.01).When using first antibody-free renal sections as negative control,expression of VIM was significantly higher than those in group S(P0.01).The indexes mentioned above were obviously lower in group T than those in group U(P0.05).TDI,TFI,and TICI positively correlated with expression of VIM respectively.Conclusions: VIM probably participates in the promoting of inflammatory reaction and the developing course of RIF;Tanshinone could inhibit the expression of VIM in renal tubulointerstitial,which may relieve RIF to some extent.
