癫痫灶中凋亡调控基因Bcl-2和Bax的表达

摘    要:目的研究难治性癫痫患者癞痫灶脑组织中Bcl-2、Bax的表达情况、以探讨癫痫与细胞凋亡调节基因之间的关系。方法采用免疫组化染色法(二步法)检测30例癫痫灶及10例周边脑组织中Bcl-2、Bax的表达,并进行统计学处理。结果Bcl-2、Bax在癫痫灶及周边脑组织均有一定的表达、癫痫灶中Bcl-2的表达明显增多(P=0.003):而癫痫灶及周边脑组织中Bax的表达比较差别无统计学意义(P=0.198)。结论 癫痫灶中存在细胞凋亡现象,且Bcl-2基因家族对此细胞凋亡起重要调控作用,Bax的表达与周边脑组织比较,差别无统计学意义、估计其作用有其他因素参与。

Abstract:Objective To study the relationship between the mechanism of epilepsy and the regulatory genes of apoptosis by analyzing the expressions of Bcl-2 and Bax in epileptogenic foci of intractable epilepsy patients. Methods The expressions of Bcl-2 and Bax in the epileptogenic foci in 30 cases and the surrounding brain tissues of 10 cases were determined by immunohistochemical staining (the two-step method) and the data were analyzed statistically. Results Bcl-2 and Bax were expressed both in the epileptogenic foci and surrounding brain tissues. The expression of Bcl-2 in the epileptogenic foci was obviously enhanced in comparison with that in the surrounding brain tissues (P=0.003) while the expression of Bax showed no statistical difference between in the epileptogenic foci and in the surrounding brain tissues (P=0.198). Conclusion The phenomenon of apoptosis can be observed in the epileptogenic foci. The Bcl-2 family poses important influence on regulating and controlling over the apoptosis. The expression of Bax in the epileptogenic foci shows no statistical difference in comparison with that in the surrounding brain tissues due to the possible participation of some other factors.
