胃与大肠腺癌中CXCL12和CXCR4蛋白的表达及相关性

摘    要:目的:研究趋化因子(C-X-C chemokine ligand 12, CXCL12)及其受体(C-X-C chemokine receptor 4, CXCR4)蛋白在胃和大肠腺癌中的表达水平,探讨二者与胃和大肠腺癌发生、发展、浸润、转移的关系及临床意义。方法应用免疫组织化学方法检测胃腺癌、正常胃黏膜组织和大肠腺癌、正常大肠黏膜组织中CXCL12、CXCR4蛋白的表达情况,对其表达水平及其与临床病理的相关性做统计学分析。结果胃腺癌组织中CXCL12、CXCR4蛋白的阳性表达率均高于正常胃黏膜组织,差异均有统计学意义(P均<0.01);且二者的阳性表达为有淋巴结转移组高于无淋巴结转移组,侵达浆膜层组高于侵达肌层组,差异均有统计学意义(P均<0.05)。大肠腺癌组织中CXCL12、CXCR4蛋白的阳性表达率均高于正常大肠黏膜组织,差异均有统计学意义(P均<0.01);且二者的阳性表达为有淋巴结转移组高于无淋巴结转移组,侵达浆膜层组高于侵达肌层组,差异均有统计学意义(P均<0.05)。结论 CXCL12和CXCR4在胃和大肠腺癌中高表达,可能与胃和大肠腺癌的发生、浸润、转移密切相关。

The expressions and correlations of CXCL12 and CXCR4 in gastrointestinal adenocarcinoma

Abstract:Objective To detect the expressions of CXCL12 and its receptor CXCR4 in gastrointesti-nal adenocarcinoma, and to evaluate their roles in carcinogenesis, development, infiltration and metastasis as well as their clinical significance. Methods The expressions of CXCL12 and CXCR4 protein were detected by SP immunohistochemical method in different tissues. The expression condition and there correlation with clini-cal pathology were analyzed statistically. Results The positive expressions of CXCL12 and CXCR4 in gastric adenocarcinoma tissue both were higher than those of the normal gastric mucosa tissue , and the differences all had statistical significance(Pall〈0.01). The positive expressions of CXCL12 and CXCR4 were closely correlat-ed with metastasis of lymph node and depth of tumor infiltration in gastric adenocarcinoma group , and the dif-ferences all had statistical significance (Pall〈0.05). The expressions of CXCL12 and CXCR4 proteins in the colorectal adenocarcinoma tissue were obviously higher than those of the normal colorectal mucosa tissue , and the differences all had statistical significance (Pall〈0.01). The positive expressions of CXCL12 and CXCR4 were closely correlated with depth of tumor infiltration and metastasis of lymph node in colorectal adenocarci-noma group, and the differences all had statistical significance(Pall〈0.05). Conclusion CXCL12 and its re-ceptor CXCR4 may play an important role in tumorigenesis progression , infiltration and metastasis of gastroin-testinal carcinoma.
