人脑胶质母细胞瘤细胞系SHG-44中血管生成因子和细胞周期调控因子的表达

摘    要:目的探讨人脑胶质母细胞瘤细胞系SHG44长期传代后的某些生物学特性和免疫表型特征。方法采用免疫组织化学和原位杂交技术对SHG44细胞增殖活性、中间丝蛋白共存、肿瘤蛋白、血管生成因子和细胞周期调控相关蛋白表达水平进行检测。结果传代130~150代的SHG44细胞呈胶质纤维酸性蛋白弱阳性而波形蛋白强阳性;Ki67、增殖细胞核抗原平均标记指数分别为83.5%±10.2%、70.0%±18.7%。p21ras、cerbB2、表皮生长因子及其受体、碱性成纤维细胞生长因子及其受体FGFR均显过度表达。血管内皮生长因子(VEGF)及VEGFmRNA的表达情况亦相似。该细胞还具有诱导型一氧化氮合酶表达和p16、p53、周期素依赖性激酶4、cyclinD1蛋白积聚(平均标记指数分别为43.1%±11.2%、20.7%±6.6%、33.1%±11.4%和29.8%±4.7%)。结论SHG44细胞存在上述生长因子、肿瘤蛋白表达异常和细胞周期的失控,使SHG44细胞生长旺盛,增殖活性高,并且血管生成能力强,具有很高的恶性表型。

Abstract:Objective To investigate the biological features and immunophenotypes of human glioblastoma cell line SHG44 after long term passage. Methods Immunohistochemistry and in situ hybridization were used to study the proliferative activity, intermediate filament protein coexistance, expressions of oncoprotein, angiogenic factors and cell cycle regulation factors. Results After 130 to 150 passages, SHG44 cells were weakly positive for glial fibrillary acidic protein (GFAP), but strongly positive for vimentin. The labeling index of Ki67 and PCNA were 83.5.2% and 70.0.7% respectively. Overexpression of p21 ras, cerbB2, epidermal growth factor (EGF) and EGF receptor were obtained. Basic fibroblast growth factor (bFGF), FGF receptor, vascular endothelial growth factor (VEGF) and inducible nitric oxide synthase (iNOS) were also upregulated in this cell line. p16, p53, cdk4 and cyclin D1 could be detected in the cells and their indices were 43.12%, 20.7%6.6%, 33.1.4% and 29.2%4.7% respectively. Conclusion Expressive abnormalities of these growth factors, their receptors and the above oncoproteins as well as disorders of cell cycle regulation contribute to the rapid growth and high degree of malignancy of this cell line.
