VEGF对人血单核细胞基质金属蛋白酶表达及活性的影响

目的 :探讨血管内皮生长因子 （VEGF）对人单核细胞源巨噬细胞 （HMDM）基质金属蛋白酶 （MMPs）的表达及其活性的影响。方法 :分别应用 RT- PCR和 Western blot检测 MMPs（MMP- 2和 MMP- 9）基因和蛋白表达 ,Zym ography活性分析检测 MMPs活性。结果 :VEGF可明显增加人单核细胞源巨噬细胞 （HMDM） MMP- 2、MMP- 9m RNA及其蛋白表达和活性。结论 :VEGF可刺激 HMDM MMP- 2、MMP- 9m RNA及其蛋白的表达 ,增强其活性 ,因而有可能促进动脉粥样硬化斑块内基质降解 ,进而引起粥样硬化斑块破裂。

Effect of VEGF on expression and activity of MMPs of human monocytes

AIM: To observe the effect of VEGF on expression and activitiy of Matrix Metalloproteinase(MMPs). METHODS: The gene and protein expression of MMPs(MMP-2 and MMP-9) were respectively detected by RT-PCR and Western blot. MMPs' activity was detected by Zymography. RESULTS: The expressions and activities of MMP-2 and MMP-9 mRNA of human monocytes were increased significantly by VEGF. CONCLUSION : As VEGF can stimulate the expressions and activities of MMP-2 and MMP-9 of human monocytes, it may increase the breakdown of matrix in lesions of atherosclerosis and enhance the lesions' vulnerability.
