丁基苯酞对异丙肾上腺素诱导大鼠心肌损伤及凋亡的作用

摘    要:目的 探讨丁基苯酞对异丙肾上腺素诱导大鼠心肌损伤保护作用及抗凋亡机制.方法 SD大鼠48只,随机分为对照组(Control组,n=12)、异丙肾上腺素组(ISO组,n=12)、异丙肾上腺素+Tween-80(丁苯酞溶媒)组(n=12)和异丙肾上腺素+丁苯酞组(n=12).7d后处死大鼠,测定血清CK-MB活性,TUNEL检测心肌组织细胞凋亡及Western Blot检测大鼠心肌caspase-3表达.结果 ISO组成功制备了心肌损伤模型.与Control 组相比,ISO组血清CK-MB活性明显增加,凋亡的心肌细胞增加,caspase-3的表达增加.丁基苯酞(NBP)能显著减轻异丙肾上腺素导致的心肌损害.与ISO组相比,ISO+NBP组血清CK-MB活性降低,差异有统计学意义(P<0.05),凋亡的心肌细胞减少,caspase-3的表达减少.结论 丁基苯酞对异丙肾上腺素引起的大鼠损伤心肌有保护作用,机制可能与其抗凋亡作用有关.

The effect of L-3-n-butylphthalide on myocardial injury and apoptosis from isoproterenol-induced rat

Abstract:Objective To investigate the effect and mechanism of dl-3-n-butylphthalide (NBP) on myocardial from isoproterenol-induced rat. Methods 48 health SD rats were randomly divided into into four groups.Control group (Control, n = 12), isoproterenol intraperitoneal injection group (ISO, n = 12), Iso +Tween group (NBP solvent)(Tween,n=12)and Iso+NBP group (NBP,n=12). After 7 days, all rats were executed. Serums were collected to detect CK-MB activity levels.TUNEL staining was used to detect myocardial cell apoptosis.Meanwhile,we furthermore measured the protein expression of caspase-3. Results The group ISO had successfully prepared the myocardial injury model.Compared with the group Control,the group ISO im- proved serum CK-MB activity, TUNEL detection of apoptotic myocardial cell increased significantly,the in- crease in caspase-3 expression by Western blot. Compared with the group ISO,the group NBP reduced serum CK-MB activity (P〈0.05),reduced the number of TUNEL detection of apoptosis in myocardial cells,and cas- pase-3 expression was decreased by Western blot. The indicators were not statistically different between group ISO and group Tween. Conclusion NBP shows protective effect in the experimental myocardial Is- ehemia injury treated .The mechanism maybe related to its resistance to apoptosis.
