摘    要:目的研制并筛选适合从腐败材料分离鼠疫菌的选择敏感培养基,对其性能进行评价。方法以自制赫氏消化液为基础,加入刺激鼠疫菌和抑制杂菌生长的生化试剂配制成2种选择敏感培养基,以常见致病菌株、耶尔森氏菌属中与鼠疫菌近源的假结核耶尔森菌、小肠结肠炎耶尔森菌评估2种培养基的敏感性和特异性;应用以上培养基分离野外腐败动物材料的鼠疫菌,评价几种选择培养基的应用效果。结果1号培养基(4%十二烷基硫酸钠选择培养基)对鼠疫菌检测的特异性高、敏感性强,培养24 h鼠疫菌发育为典型成熟菌落,应用该培养基从腐败动物材料中分离鼠疫菌阳性检出率与龙胆紫选择培养基一致。结论4%十二烷基硫酸钠选择培养基分离鼠疫菌效果优于龙胆紫选择培养基,可用于野外腐败动物材料的鼠疫菌快速检验。

Abstract:This study developed and screened a selective sensitive medium suitable for isolating Yersinia pestis from corrupted materials and evaluated its performance. We added biochemical reagents and bacteriostatic agents to Hector’s digestive juice made in our laboratory to prepare two types of selective sensitive media for stimulating Y. pestis growth. The sensitivity and specificity of the culture media were evaluated with the common pathogenic strains Yersinia pseudotuberculosis and Yersinia enterocolitica, which are closely related to Y. pestis. The above media were then used to isolate Y. pestis from corrupted animal materials in the wild, and the effects of their application were evaluated. No. 1 medium (4% sodium lauryl sulfate selective medium) had high specificity and sensitivity for the detection of Y. pestis, which formed typical mature colonies after 24 hours of culture. The positive detection rate of Y. pestis isolated from spoiled animal materials with 4% sodium lauryl sulfate selective medium was consistent with that with gentian violet selective medium. The 4% sodium lauryl sulfate selective medium performed better than the gentian violet selective medium in the isolation of Y. pestis and thus can be used for rapid detection of this pathogen in corrupted wild animal materials.
