摘    要:目的了解贵州省2013-2021年羊种布鲁氏菌的分子流行病学特征,为贵州省布鲁氏菌病疫情防控提供科学依据。方法以贵州省疾病预防控制中心2013-2021年分离、鉴定和保存的78株羊种布鲁氏菌分离株为研究对象,运用BCSP31-PCR及AMOS-PCR技术对分离株分别进行布鲁氏菌属及种的鉴定,运用多位点可变数目串联重复序列分析(MLVA-16)技术进行分型分析。对MLVA分型结果进行整理及聚类分析,了解贵州省羊种布鲁氏菌的MLVA型别分布及分子流行病学特征。结果78株羊种菌分离株经BCSP31-PCR技术均鉴定为布鲁氏菌属细菌,AMOS-PCR技术将其均鉴定为羊种布鲁氏菌,MLVA-8分型技术将其分为5种MLVA型,MLVA-16分型技术将其分为46种MLVA型,其中遵义市的MLVA-16基因型型别分布最多。基于MLVA-16分型数据聚类分析显示,78株分离株亲缘关系较近,被聚类为4个群;最小生成树图显示,78株分离株被分为8个遗传复合体和14个单体。结论贵州省羊种布鲁氏菌MLVA型别呈多样性,提示贵州省布病疫情的病原体可能存在多条传播链。

Abstract:The molecular epidemiological characteristics of Brucella melitensis (B. melitensis) in Guizhou Province from 2013 to 2021 were studied to provide a scientific basis for the prevention and control of brucellosis in Guizhou Province. A total of 78 isolates were identified as Brucella spp. strains with the BCSP31-PCR technique, and all were identified as B. melitensis with the AMOS-PCR technique. For MLVA typing based on MLVA-8 and MLVA-16 data, 78 isolates yielded 5 and 46 MLVA genotypes, respectively; the largest distribution of genotypes was observed for MLVA-16 in the Zunyi region. Cluster analysis based on MLVA-16 typing data indicated that the 78 isolates were closely related and clustered into four groups, and the minimum spanning tree mapping demonstrated that the 78 isolates were divided into 8 genetic complexes and 14 monomers. On this basis, we concluded that B. melitensis in Guizhou Province has MLVA type diversity, and multiple transmission chains of the pathogen might have caused the outbreak of brucellosis in Guizhou Province.
