趋化性细胞因子受体CXCR4与肺癌侵袭和转移的关系

目的 探讨趋化性细胞因子受体CXCR4在肺癌中的表达及其意义.方法 收集72例非小细胞肺癌患者手术标本,用免疫组织化学方法检测肿瘤组织中CXCR4的表达和微血管密度,分析CXCR4的表达水平与临床病理特征和预后的关系,12例正常肺组织作为对照.结果 72例非小细胞肺癌组织中,46例CXCR4蛋白表达阳性,阳性率为63.9%;12例正常肺组织中,仅2例CXCR4弱阳性表达,阳性率为16.7%,两者比较,差异有显著性意义(P＜0.05).肺癌组织CXCR4阳性表达与N分期、微血管密度、复发/转移和生存时间密切相关(均P＜0.05).结论 CXCR4可能促进肺癌的侵袭、转移以及血管生成并影响其预后.

Effect of Chemokine Receptor CXCR4 on the Invasion and Metastasis Capability in Lung Cancer

Objective To investigate the expression of chemokine receptor CXCR4 in non-small cell lung cancer(NSCLC) and the probable significance.Methods Tumor tissues were collected from 72 patients with NSCLC who underwent surgery from January 1996 to June 1998 at the Department of Surgery,Tongji Hospital.Twelve samples of normal lung tissues were collected as controls during the same period.Immunohistochemical study for detection of CXCR4 expression and microvessel density was performed.The expression levels of CXCR4 in tumor tissues and normal tissues were compared and the correlation between the expression levels of CXCR4 and clinicopathological factors was evaluated.Results Of 72 cancer tissue samples,46 were positive for CXCR4 and the positive rate was 63.9%.Of 12 normal tissue samples,only 2 cases were weakly positive and the positive rate was 16.7%.The cases positive for the CXCR4 expression had higher lymph node stage and microvessel density,were easier to develop recurrence or metastasis and had shorter survival time than those with negative CXCR4 expression(P<0.05).Conclusion CXCR4 over-expression in NSCLC is associated with lymph node status,microvessel density,recurrence and survival time,and CXCR4 may play an important role in the process of invasion,metastasis and angiogenesis,thus affecting the prognosis of NSCLC.
