摘 要: | 目的探讨北虫草提取物抗衰老的作用,为新资源食品的有效利用提供科学依据。方法昆明种小鼠70只,随机分为对照组、衰老模型组和3个剂量北虫草提取物组(3.3、10、30 g/kg)。建立D-半乳糖致小鼠亚急性衰老模型,以称质量法测定胸腺和脾指数,对-二甲氨基苯甲醛显色法测定小鼠皮肤中羟脯氨酸含量,荧光分光光度法测定肝组织中脂褐质含量。结果北虫草提取物低剂量组的胸腺指数和脾指数分别为51.3±4.5和27.6±3.6,明显高于模型组(分别为45.6±4.8和23.6±3.6)(P0.05,P0.01);中、高剂量组的上述2个指数也同样明显高于模型组(P0.05,P0.01)。北虫草提取物3个剂量组皮肤羟脯氨酸的含量均明显高于模型组(P0.05,P0.01),北虫草小分子肽低、中2个剂量组肝组织中脂褐质的含量明显低于模型组(P0.01)。结论北虫草提取物能促进衰老模型小鼠的皮肤羟脯氨酸生成,降低其肝组织脂褐质含量,并提高其胸腺指数和脾指数,提示北虫草提取物可能具有抗衰老的生物作用。

Abstract: | Objective: To study the antiageing effect of Cordyceps extract, in order to provide the scientific basis for effective use of new resources of food. Methods Seventy mice of Kun-Ming species were randomly divided into 5 groups : control, subacute ageing model, and 3 doses of Cordyceps extract (3.3,10.0,30.0 g/kg for 6 weeks). Subacute ageing model was induced by D-galactose. The index of thymus and spleen were determined by weighing method, the content of skin hydroxyproline by para-2,4-amino benzaldehyde coloration method, and the content of liver lipofuscin by fluorescence spectrophotometry. Results The index of thymus and spleen in low dose of Cordyceps group(51.3±4.5,27.6±3.6 respeetivly) was significantly higher than in model group (45.6±4.8, 23.6±3.6 respectively) , (P〈0. 05 ,P〈0.01 ). Those in middle and high dose group were also higher than in model group( P〈0.05 ,P〈0.01 ). The skin hydroxyproline content in all 3 doses of Cordyceps groups was higher than in model group, and the liver ]ipofuscin in low and middle dose of Cordyceps group was lower than in model group (P 〈0.01 ). Conclusion Cordyceps extract could promote skin hydroxyproline formation in simulated ageing mice, decrease their liver lipofuscin content, and increase their thymus index and spleen index, suggesting that Cordyceps extract may have antiageing biological function. |
