摘    要:目的:探讨GH,HGF,bFGF,TGF,IGF-Ⅰ等多种生长因子联合使用对体外培养人骨骼肌卫星细胞增殖能力的影响。方法:选取临床手术切取的儿童肌肉活检标本20例,实验分为4组:TGF+IGF-Ⅰ组(5例);bFGF+TGF-β组(5例);GH+HGF组(5例);GH,HGF,bFGF,TGF,IGF-Ⅰ组(5例)。进行体外骨骼肌卫星细胞的培养,然后在培养基中分别加GH,HGF,bFGF,TGF,IGF-Ⅰ等生长因子,采用MTT法测TGF+IGF-Ⅰ、bFGF+TGF-β、GH+HGF及GH,HGF,bFGF,TGF,IGF-Ⅰ对骨骼肌卫星细胞增殖的影响。结果:在体外培养过程中,GH,HGF,bFGF,TGF,IGF-Ⅰ联合使用促进骨骼肌卫星细胞增殖的作用明显高于TGF+IGF-Ⅰ组、bFGF+TGF-β组及GH+HGF组,差异有显著性(P<0.01);而TGF+IGF-Ⅰ组与bFGF+TGF-β组;TGF+IGF-Ⅰ组与GH+HGF;bFGF+TGF-β组与GH+HGF组骨骼肌卫星细胞的增殖能力比较,差异无显著性(P>0.05)。结论:TGF+IGF-Ⅰ组;bFGF+TGF-β组及GH+HGF组作用对体外培养大鼠骨骼肌卫星细胞有增殖作用,但其作用较弱,而GH,HGF,bFGF,TGF,IGF-Ⅰ联合使用对骨骼肌卫星细胞增殖作用明显。本实验为体外对骨骼肌卫星细胞的增殖能力提供了重要的实验依据。

Abstract:Objective: To investigate the combination effects of basic fibroblast growth factor(bFGF),transforming growth factor-beta(TGF-β),growth hormone (GH),hepatocyte growth factor(HGF),insulin-like growth factor(IGF-Ⅰ) on the proliferation of human skeletal muscle satellite cells in vitro.Methods: Twenty specimens of skeletal muscle were divided equally into four groups as TGF+IGF-Ⅰ group,bFGF+TGF-β group,GH+HGF group and GH+HGF+bFGF+TGF+IGF-Ⅰ group according to the medium with different growth factors.MTT method was used to study the proliferation of the skeletal muscle satellite cells.Results: The cell proliferation ability were lower in bFGF+TGF-β group,TGF+IGF-Ⅰ group and GH+HGF group than in GH+HGF+bFGF+TGF+IGF-Ⅰ group(P<0.01).Conclusion: The combination of GH,HGF,bFGF,TGF and IGF-Ⅰ could has a superior effect on the proliferation of human skeletal muscle satellite cells in vitro.
