氧化苦参碱大鼠肠道吸收机理及吸收部位的研究

摘    要:目的探明氧化苦参碱在大鼠肠道各区段的吸收动力学特征、吸收部位及吸收机制,以期对药物长效缓释制剂的设计提供重要依据。方法采用大鼠在体肠灌流吸收实验,研究了氧化苦参碱的吸收部位和吸收动力学特征。结果氧化苦参碱在肠道的吸收呈现一级吸收动力学特征且吸收机制为被动转运。药物在小肠部位吸收良好,吸收速度按空肠、回肠、十二指肠、结肠的顺序依次下降,吸收速率常数依次为0.237、0.225、0.200和0.062h-1。结论氧化苦参碱的吸收窗比较长,适于制备缓释给药系统。

Abstract:AIM: To study the absorption mechanism and absorption site of oxymatrine in rats. METHODS: The absorption kinetics and absorption site of oxymatrine were investigated using in situ perfusion method in rats. RESULTS: The absorption mechanism of oxymatrine complied with the passive diffusion and first order kinetics. The absorption rate decreased according to the turn of jejunum, ileum, duodenum and colon. The absorption rate constants were 0.237 , 0.225 , 0.200 and 0.062 h -1 at jejunum, ileum, duodenum and colon, respectively. CONCLUSION: Oxymatrine can be prepared and processed into sustained-release dosages forms.
