恒磁场对人脐静脉内皮细胞一氧化氮合酶活性的影响

摘    要:目的:观察恒磁场(staticmagneticfield,SMF)对培养的人脐静脉内皮细胞(humanumbilicalveinendotheliumcell,HUVEC)一氧化氮合酶(nitricoxidesynthase,NOS)活性表达的影响,以探讨SMF能否用于经皮腔内冠状动脉成形术(percutaneoustransluminalcoronaryangioplasty,PTCA)及冠脉内支架植入术后冠脉再狭窄(restenosis,RS)的防治。方法:用含100g/L小牛血清的DMEM培养液体外培养人脐静脉内皮细胞(ECV304),分别在给予SMF和无SMF条件下培养,行ABC免疫酶染色法,采用图像分析法分析SMF对HUVEC的NOS活性的影响。结果:实验组内皮细胞结构型NOS(endotheliumconstructiveNOS,ecNOS)表达较对照组增强,差异显著(P<0.01),实验组与对照组诱生型NOS(inducedNOS,iNOS)均无表达。结论:恒磁场增强了培养的HUVEC的ecNOS的表达;恒磁场对PTCA支架植入术后的冠脉再狭窄可能具有防治作用。

Abstract:AIM:This study was designed to evaluate the effect of static magnetic field (SMF) on the expression of nitric oxide synthase (NOS) in human umbilical vein endothelial cells(HUVECs) and to discuss if SMF can be used to treat the coronary artery restenosis after percutanuous transluminal coronary angioplasty (PTCA) and in stent. METHODS:Human umbilical vein endothelial cells line (ECV304) were cultured in vitro with DMEM and 10 percent calf plasma. The HUVECs were cultured with SMF and without SMF respectively and ABC immunohistochemistry technique and image analyzing method were used to evaluate the effect of the CMF on the expression of NOS. RESULTS:The expression of ecNOS in experimental group was more than the control group (P< 0.01), There was no expression of iNOS in each group. CONCLUSION:SMF can enhance the expression of ecNOS in cultured HUVECs and may be suitable in the prevention of the restenosis after PTCA.
