高效液相色谱法同时测定二十五味珍珠丸中7种成分及其特征图谱

目的：建立二十五味珍珠丸7种成分含量测定方法及HPLC特征图谱，为质量控制提供依据。方法：Waters Symmetry-C₁₈色谱柱（4.6 mm×250 mm，5 μm），以乙腈和0.1%磷酸溶液为流动相，梯度洗脱，检测波长：280 nm（用于特征图谱建立和丁香酚含量测定）、290 nm（桂皮醛）、274 nm（去氢二异丁香酚）、343 nm（胡椒碱）、225 nm（木香烃内酯与去氢木香内酯）、272 nm（没食子酸）。结果：对二十五味珍珠丸7个有效成分进行了含量测定；同时建立了特征图谱并对特征峰进行指认。结论：建立的含量测定方法及HPLC特征图谱符合相关要求，适合二十五味珍珠丸的质量控制。

Simultaneous determination of seven components and typical chromatogram of Ershiwuwei Zhenzhu Pills by HPLC

OBJECTIVE To establish a new method for quality control and typical HPLC chromatogram for Ershiwuwei Zhenzhu Pills. METHODS HPLC was used on a Waters Symmetry-C₁₈ column (4.6 mm×250 mm, 5 μm) and diode array detector with gradient elution solvent system composed of acetonitrile-0.1% phosphoric acid solution. Detection wavelengths were 280 nm (typical chromatogram and eugenol), 290 nm (cinnamaldehyde), 274 nm (dehydrodiisoeugenol), 343nm (piperine), 225 nm (costunolide and dehydrocostus lactone) and 272 nm (gallic acid). RESULTS Seven effective components were determined in Ershiwuwei Zhenzhu Pills. Typical HPLC chromatogram was established and five characteristic peaks were identified. CONCLUSION The established method for quantitative determination and the typical HPLC chromatogram have desirable precision, reproducibility and stability, and can be applied for quality control of Ershiwuwei Zhenzhu Pills.
