慢性牙周炎患者和健康人牙龈成纤维细胞细胞活性和凋亡的比较研究

目的:探索慢性牙周炎患者和健康人牙龈成纤维细胞(HGFs)细胞活性和凋亡之间的差异性,为阐明牙周炎的易感性提供实验依据。方法:收集慢性牙周炎患者和健康人的健康牙龈,进行体外培养获得HGFs,采用MTT法比较两组细胞的细胞活性,实时荧光定量PCR检测抗凋亡基因Bcl-2和促凋亡基因Bax在两种细胞中的表达水平及其比值。结果:与健康组相比,慢性牙周炎组HGFs细胞活性在培养4 d后明显下降(P＜0.05),而且在体外培养2 d、4 d细胞中Bcl-2/Bax的比值下降(P＜0.05),具有时间依赖性。结论:慢性牙周炎患者HGFs细胞活性下降,细胞凋亡增强,从细胞水平说明其存在牙周炎易感性。

Comparison of Viability and Apoptosis of Gingival Fibroblasts in the Patients with Chronic Periodontitis and Healthy People.

Objective: To investigate the differences in the proliferation and apoptosis of human gingival fibroblasts (HGFs) between patients with chronic periodontitis and healthy people and provide experimental evidence for the susceptibility of chronic periodontitis. Methods: HGFs were obtained from patients with chronic periodontitis and healthy people and cultured in vitro. Cell activity between two groups was detected by MTT assay. Real-time PCR was used to detect the expression levels of anti-apoptotic gene Bcl-2 and pro-apoptotic gene Bax in HGFs from the two groups, and the ratio of Bcl-2 to Bax was compared. Results: Compared with the healthy group, the cell activity of the HGFs in chronic periodontitis group decreased significantly after 4 days culture (P<0.05). The ratio of Bcl-2/Bax significantly decreased after 2 days and 4 days in vitro in a time-dependent manner (P<0.05). Conclusion: HGFs cells from patients with chronic periodontitis show low cell activity and high cell apoptosis, which indicates the susceptibility of periodontitis.
