神经性痛和炎性痛大鼠伏核内CGRP mRNA表达的变化

摘    要:为了研究神经性痛和炎性痛大鼠伏核内CGRP mRNA表达的变化,本研究分别制备神经性痛及炎性痛大鼠动物模型,并运用实时定量逆转录聚合酶链式反应(PCR)技术,观察大鼠伏核内CGRP mRNA含量的改变。实验中以β-actin为内参,记录大鼠伏核内CGRP mRNA表达产物的CT值,并计算每一样品对应其内参的相对含量,分析随时间变化致痛因素对大鼠伏核内CGRP mRNA表达产物含量的影响。结果显示:神经损伤组5、10、20、30d及炎症组3d,CGRP mRNA的表达比正常对照组明显增高(P<0.05);而在炎症组7d时,CGRP mRNA的表达与对照组相比无显著性差异(P>0.05)。以上结果提示CGRP作为一种神经递质或调质参与痛觉的调制过程,其主要是在炎症早期发挥作用。

Abstract:To examine the changes of CGRP mRNA expression in the nucleus accumbens of rats with neuropathic and inflammatory pain, the neuropathic and the inflammatory pain models in rats were prepared in the present study. The real time quantitative RT-PCR technique was used for CGRP mRNA recording and detected the changes of CGRP mRNA. With β-actin as an endogenous control, CT value of CGRP mRNA in the nucleus accumbens of rats was recorded and the relative amount of every sample against its endogenous control was calculated, the influence of pain on CGRP mRNA eypression in different times was analyzed. The results showed that on 5, 10, 20, 30 days in nerve ligation group and on 3 days in the inflammatory group, the expression of CGRP mRNA were more higher than that in the intact rats (P<0.05). But on 7 days in the inflammatory group, there was no significant difference in the expression of CGRP with the control group (P>0.05). The results indicate that CGRP may be involved in the pain modulation as a kind of neurotransmitter or modulator, but it exerts its effect mainly in the early stage of inflammation.
