一个AB型开米拉家系遗传状态的研究

摘    要:为了研究罕见开米拉血型家系的基因遗传状态,对先证者家系部分样本进行ABO基因序列测定,用流式反向序列特异性寡核苷酸探针方法测定HLA-A、B、DRB1基因位点,用PCR-序列特异性引物法测定HLA-A、B、DRB1基因位点,对16个短串联重复片段位点进行荧光标记复合扩增。结果发现:A3B3型家系的2个体中,ABO基因、HLA-B、DRB1基因、多个STR位点出现了2个以上的等位基因。结论:利用基因分型技术分析开米拉血型能清楚地提示此罕见血型的遗传状态,增进了对此遗传方式的认识。

Abstract:In order to study the genetic status of a rare chimeric family, some samples of A3B3 family were identified by sequencing of ABO gene; flow-rSSO and PCR-SSP were used to detect loci of HLA-A,B,DRB1 genes, and multiplex amplifying with fluorescence-dye were performed for 16 short tandem repeat (STR) loci. The results indicated that two individuals from A3B3 family contained more than two alleles at ABO gene, HLA-B,DRB1 and some STR loci. In conclusion, analysis of chimeric blood group by using genotyping techniques clearly demonstrating genetic status of this rare chimeric blood group promotes further elucidation of the existing state of specific genetic status.
