摘    要:目的 探讨甲状腺激素(TH)对新生大鼠脑血管发育的影响.方法 取18只清洁级SD成年雌性大鼠与3只清洁级SD雄性大鼠交配后随机将孕鼠分入3组:丙基硫氧嘧啶(PTU)干预组、TH干预组和对照组.待每组仔鼠产后第1天即采用电化学发光法检测其血清总三碘甲状腺原氨酸(TT3)、总甲状腺素(TT4)水平;采用酶联免疫吸附试验法测仔鼠脑组织血管内皮细胞生长因子(VEGF)水平.应用SPSS 16.0软件进行统计学分析.结果 TH干预组仔鼠血清TT4(103.50±2.37) μg·L-1]、TT3(1 269.63±32.18) ng·L-1]与脑组织VEGF水平(53.46±10.62) μg·L-1]均高于对照组TT4 (45.11±0.43) μg·L-1,TT3 (844.57± 20.85) ng·L-1,VEGF (11.15±1.33) μg·L-1](Pa<0.001).同时,PTU干预组新生鼠以上各指标TT4 (16.83±0.45) μg·L-1,TT3 (142.35±7.76) ng·L-1,VEGF(8.69±0.48) μg·L-1]均低于对照组(Pa<0.001).结论 TH干预可增加仔鼠脑组织VEGF水平,对脑血管的发育有促进作用.

Abstract:Objective To investigate the effect of thyroid hormone(TH) on brain angiogenesis in newborn rats.Methods Eighteen mature virgin females and 3 mature males were selected.Females of each nest were divided into 3 groups randomly after mating:6-N-propyl-2-thiouracil(PTU) regulated group,thyroid hormone(TH) regulated group and control group.Newborns of post neonatal day 1st in each group were used:their serum total thyroxine(TT4),total triiodothyronine(TT3) were estimated by electrochemical luminescence and brain vascular endothelial growth factor(VEGF) were measured by enzyme-linked immunosorbent assay.SPSS 16.0 software was used to analyze the data.Results Serum TT4,TT3,brain VEGF in control groupTT4(45.11±0.43) μg·L-1,TT3(844.57±20.85) ng·L-1,VEGF(11.15±1.33) μg·L-1] were decreased significantly than those in TH regulated group TT4(103.50±2.37) μg·L-1,TT3(1 269.63±32.18) ng·L-1,VEGF(53.46±10.62) μg·L-1](Pa<0.001).However,the levels of serum TT4,TT3 and brain VEGF in PTU regulated group TT4(16.83±0.45) μg·L-1,TT3(142.35±7.76) ng·L-1,VEGF(8.69±0.48) μg·L-1] were decreased significantly compared with those in control group.Conclusions Regulated TH can increase the level of brain VEGF in newborn rats.TH has a substantial impact on vasculature development in the brain of newborn rats.
