宫颈病变中端粒酶活性与高危HPV感染关系的研究

摘    要:目的探讨端粒酶活性与高危HPV感染在宫颈病变中的表达情况及其相互关系。方法选择宫颈癌、CIN各级及慢性宫颈炎组织标本70例。应用TRAP—PCR银染定性检测端粒酶活性,并进行高危HPVDNA—PCR定性检测。结果随着宫颈病变级别的增强端粒酶阳性表达率及高危HPV阳性表达率增加。结论端粒酶活性表达在子宫颈癌的发生发展中可能起重要的作用;在宫颈癌发生发展过程中端粒酶活性与高危型HPV感染密切相关,宫颈高危型HPV感染可能通过激活端粒酶诱发癌变。

Abstract:Objective To study telomerase activity in cervical intraepithelial neoplasia(CIN),and to assess whether it was activated during cervical malignant transformation and to look for a possible association with high risk human papillomavirus(HR-HPV)infection.Methods Histologically confirmed invasive cervical carcinoma,intraepithelial neoplasia(CIN),and benign cervices were assayed for telomerase activity using a commercial telomerase polymerase chain reaction(PCR)-TRAP(telomere repeat amplification protocol assay).The same cases were subjected to PCR detection of HR-HPV using type specific.Result Expression of telomerase activity and HR-HPV appeared in cervical cancer and cervical intraepithelial neoplasm.Conclusions Expression of telomerase activity appears in cervical cancer and cervical intraepithelial neoplasm,and is closely related to the occurrence and progress of cervical cancers.There seems to be a clear association between telomerase activity and HR-HPV status in the development of cervical cancer.HR-HPV may cause carcinomatous change by activating telomerase.
