脑动脉硬化大鼠模型的试制及药物干预

摘    要:目的研究建立脑动脉硬化症的大鼠模型并观察桂利嗪及中药健脑颗粒的治疗作用。方法用高血压合并高血脂方法建立脑动脉硬化大鼠模型,分别给予桂利嗪及中药健脑颗粒治疗,于术后15周检测血压、血清脂质水平、血浆内皮素(ET)、一氧化氮(NO)、血栓素B2(TXB2)及6-酮-前列腺素F1α(6-Keto-PGF1α)含量,并对脑血管及脑组织作病理检查。结果模型组大鼠血压、血清脂质水平、ET/NO、TXB2/6-Keto-PGF1α比值均明显上升(P〈0.01);脑动脉内膜内皮细胞呈广泛性变性、坏死、脱落,粥样斑块形成。经给药治疗后上述指标均有一定改善。结论采用高血压合并高血脂方法可建立与人类疾病相似的实验性脑动脉硬化大鼠模型,且药物治疗具有可逆性。

Abstract:Objective To establish the model of cerebral arteriosclerosis in rats and to observe the effects of tablet apotomin and Jiannao granule. Methods The model of cerebral arteriosclerosis in rats was established with both hypertension and hyperlipidemia. Tablet apotomin and Jiannao Granule were given respectively for therapy. The blood pressure, the level of serum lipid ,and the levels of ET, NO ,TXB2 and 6-Keto-PGF1a in plasma were detected 15 weeks after operation. Cerebral tissue and vascular pathological observation was performed. Results The blood pressure, ET/NO, TXB2/6-Keto-PGF1a were significantly increased in the rats of model group than those in sham operation group (P 〈 0. 01 ). Cerebral endarterium endothelial cells showed degeneration, necrosis and ablation extensively. Atherosclerosis plaque was formed. The above parameters were significantly improved in both medication groups after receiving treatment. Conclusions The experimental model of cerebral arteriosclerosis in rats could be established successfully with both hypertension and hyperlipidemia, and it can be reversed by medication.
